The innate immune response to human immunodeficiency virus-1 infection

The data presented in this dissertation demonstrate that the innate immune system in the thymus recognizes and responds to HIV-1 infection. We illustrate that the plasmacytoid dendritic cells (p DC) are the key mediators of this response. The primary role of the p DC is to secrete interferon-alpha (IFN-alpha) to warn cells of the presence of a pathogen. The local secretion of IFN-alpha results in the upregulation of interferon stimulated genes (ISG) that inhibit viral replication. We utilized the expression of an ISG, Myxovirus resistance A (Mx A), as a surrogate for the secretion of IFN-alpha as Mx A is only expressed when cells are stimulated via the IFN-receptor.;The data shown here suggest that IFN-alpha secreted by p DC plays an… [cont.]

The second edition of Primary Immunodeficiency Diseases presents discussions of gene identification, mutation detection, and clinical and research applications for over 100 genetic immune disorders--disorders featuring an increased susceptibility to infections and, in certain conditions, an icreased rate of malignancies and autoimmune disorders. Since the publication of the first edition, a flurry of new disease entities has been defined and new treatment regimens have been introduced, the most spectacular being successful treatment by gene therapy for two genotypes of combined immunodeficiency. The first edition marked a historic turning point in the field of immunodeficiencies, demonstrating that many of the disorders of the immune… [cont.]

Herpes simplex virus (HSV) infection the causative agent for genital herpes is an important cofactor in HIV transmission. Published evidence indicates that continued transmission is facilitated by the large number of undiagnosed cases, the frequency of atypical or pauci-symptomatic disease and the occurrence of asymptomatic shedding. This book describes: i) the diagnostic advantages of PCR as a gold standard test for screening high-prevalence populations; ii) the risk factors associated with genital HSV type-1 and type-2 infections and its influence on the acquisition of genital herpes in HIV positive individuals and iii) the reconstitution of HSV-specific T-cell responses in HIV infected patients and healthy controls. Although strategies… [cont.]

Over the last 50 years, the human immunodeficiency virus (HIV) has swept across the six continents, claiming several million lives yearly. Despite sustained intense research efforts, there is still no cure for HIV. For billions of years, viruses evolved strategies to enter and take control of organisms to generate progeny viruses, including means of hijacking the cellular protein synthesis machinery. Understanding these mechanisms opens a unique window of opportunity: that of eventually being able to specifically inhibit virus protein production and thus put a stop to virus replication. In this work, HIV-1 protein synthesis was studied, uncovering the presence of an internal ribosome entry site (IRES) in the genetic code of HIV-1. IRESes… [cont.]

Immunodeficiency (or immune deficiency) is a state in which the immune system's ability to fight infectious disease is compromised or entirely absent. Most cases of immunodeficiency are acquired ("secondary") but some people are born with defects in their immune system, or primary immunodeficiency. Transplant patients take medications to suppress their immune system as an anti-rejection measure, as do some patients suffering from an over-active immune system. A person who has an immunodeficiency of any kind is said to be immunocompromised. An immunocompromised person may be particularly vulnerable to opportunistic infections, in addition to normal infections that could affect everyone.
